RESUMO

Background: The microbiome likely plays a role in tuberculosis (TB) pathogenesis. We evaluated the site-of-disease microbiome and predicted metagenome in people with presumptive tuberculous pericarditis, a major cause of mortality, and explored for the first time, the interaction between its association with C-reactive protein (CRP), a potential diagnostic biomarker and the site-of-disease microbiome in extrapulmonary TB. Methods: People with effusions requiring diagnostic pericardiocentesis (n=139) provided background sampling controls and pericardial fluid (PF) for 16S rRNA gene sequencing analysed using QIIME2 and PICRUSt2. Blood was collected to measure CRP. Results: PF from people with definite (dTB, n=91), probable (pTB, n=25), and non- (nTB, n=23) tuberculous pericarditis differed in ß-diversity. dTBs were, vs. nTBs, Mycobacterium-, Lacticigenium-, and Kocuria- enriched. Within dTBs, HIV-positives were Mycobacterium-, Bifidobacterium- , Methylobacterium- , and Leptothrix -enriched vs. HIV-negatives and HIV-positive dTBs on ART were Mycobacterium - and Bifidobacterium -depleted vs. those not on ART. Compared to nTBs, dTBs exhibited short-chain fatty acid (SCFA) and mycobacterial metabolism microbial pathway enrichment. People with additional non-pericardial involvement had differentially PF taxa (e.g., Mycobacterium -enrichment and Streptococcus -depletion associated with pulmonary infiltrates). Mycobacterium reads were in 34% (31/91), 8% (2/25) and 17% (4/23) of dTBs, pTBs, and nTBs, respectively. ß-diversity differed between patients with CRP above vs. below the median value ( Pseudomonas -depleted). There was no correlation between enriched taxa in dTBs and CRP. Conclusions: PF is compositionally distinct based on TB status, HIV (and ART) status and dTBs are enriched in SCFA-associated taxa. The clinical significance of these findings, including mycobacterial reads in nTBs and pTBs, requires evaluation.

The aim of this study was to compare the running demands of transition games (TGs) and official matches, analysing their requirements according to the performance of each position. An observational design was used to examine the activity of 20 soccer players during official matches and TGs. GPS technology was used to monitor the total distance covered (DC), distance at speeds between 14-17.9 km•h-1, 18-21 km•h-1, and above 21 km•h-1, peak speed, accelerations and decelerations above 2.5 m•s-2, and Player Load for both activities. All players were assigned to groups: centre-backs (CBs), fullbacks (FBs), defensive midfielders (DMFs), offensive midfielders (OMFs), wide midfielders (WMFs) and strikers (Ss). TGs showed greater total DC, DC 14-17.9 km•h-1, DC 18-21 km•h-1, DC >21 km•h-1, accelerations and decelerations >2.5 m•s-2, and Player Load (p < 0.01). CBs, FBs and Ss showed more DC, DC 14-17.9 km•h-1, DC 18-21 km•h-1, DC >21 km•h-1, accelerations and decelerations >2.5 m•s-2 and Player Load in TGs (p < 0.01). In the midfielder positions, transition game players showed greater DC 18-21 km•h-1, DC >21 km•h-1, accelerations and decelerations >2.5 m•s-2 than in matches (p < 0.05). DMFs showed higher total DC (p < 0.05) and WMFs greater DC and DC 14-17.9 km•h-1 (p < 0.01) in these drills. During transition games CBs showed greater DC 14-17.9 km•h-1 than FBs, and greater DC than Ss (p < 0.05). FBs performed more decelerations >2.5 m•s-2 than DMFs and OMFs (p < 0.05). TGs produced a homogenized load in soccer players, independent of their position, which exceeded the external load of official matches.

Background: Latent tuberculosis infection (LTBI) is common in people living with HIV (PLHIV) in high TB burden settings. Active TB is associated with specific stool taxa; however, little is known about the stool microbiota and LTBI, including in PLHIV. Method: Within a parent study that recruited adult females with HIV from Cape Town, South Africa into predefined age categories (18-25, 35-60 years), we characterised the stool microbiota of those with [interferon-γ release assay (IGRA)- and tuberculin skin test (TST)-positive] or without (IGRA- and TST- negative) LTBI (n=25 per group). 16S rRNA DNA sequences were analysed using QIIME2, Dirichlet Multinomial Mixtures, DESeq2 and PICRUSt2. Results: No α- or ß-diversity differences occurred by LTBI status; however, LTBI-positives were Faecalibacterium-, Blautia-, Gemmiger-, Bacteroides-enriched and Moryella-, Atopobium-, Corynebacterium-, Streptococcus-depleted. Inferred metagenome data showed LTBI-negative-enriched pathways included several involved in methylglyoxal degradation, L-arginine, putrescine, 4-aminobutanoate degradation and L-arginine and ornithine degradation. Stool from LTBI-positives demonstrated differential taxa abundance based on a quantitative response to antigen stimulation (Acidaminococcus-enrichment and Megamonas-, Alistipes-, and Paraprevotella-depletion associated with higher IGRA or TST responses, respectively). In LTBI-positives, older people had different ß-diversities than younger people whereas, in LTBI-negatives, no differences occurred across age groups. Conclusion: Amongst female PLHIV, those with LTBI had, vs. those without LTBI, Faecalibacterium, Blautia, Gemmiger, Bacteriodes-enriched, which are producers of short chain fatty acids. Taxonomic differences amongst people with LTBI occurred according to quantitative response to antigen stimulation and age. These data enhance our understanding of the microbiome's potential role in LTBI.

Background: Tuberculosis (TB), a major cause of disease and antimicrobial resistance, is spread via aerosols. Aerosols have diagnostic potential and airborne-microbes other than Mycobacterium tuberculosis complex (MTBC) may influence transmission. We evaluated whether PneumoniaCheck (PMC), a commercial aerosol collection device, captures MTBC and the aeromicrobiome of people with TB. Methods: PMC was done in sputum culture-positive people (≥30 forced coughs each, n=16) pre-treatment and PMC air reservoir (bag, corresponding to upper airways) and filter (lower airways) washes underwent Xpert MTB/RIF Ultra (Ultra) and 16S rRNA gene sequencing (sequencing also done on sputum). In a subset (n=6), PMC microbiota (bag, filter) was compared to oral washes and bronchoalveolar lavage fluid (BALF). Findings: 54% (7/13) bags and 46% (6/14) filters were Ultra-positive. Sequencing read counts and microbial diversity did not differ across bags, filters, and sputum. However, microbial composition in bags (Sphingobium-, Corynebacterium-, Novosphingobium-enriched) and filters (Mycobacterium-, Sphingobium-, Corynebacterium-enriched) each differed vs. sputum. Furthermore, sequencing only detected Mycobacterium in bags and filters but not sputum. In the subset, bag and filter microbial diversity did not differ vs. oral washes or BALF but microbial composition differed. Bags vs. BALF were Sphingobium-enriched and Mycobacterium-, Streptococcus-, and Anaerosinus-depleted (Anaerosinus also depleted in filters vs. BALF). Compared to BALF, none of the aerosol-enriched taxa were enriched in oral washes or sputum. Interpretation: PMC captures aerosols with Ultra-detectable MTBC and MTBC is more detectable in aerosols than sputum by sequencing. The aeromicrobiome is distinct from sputum, oral washes and BALF and contains differentially-enriched lower respiratory tract microbes.

The aims of this study were to determine the effect of different compositions in transition games (TGs) on the load of soccer players and to evaluate their performance in physical tests. Using a GPS system, 18 players were monitored during: 3vs2, 2vs1 and 1vs1. Distance covered (DC), DC 18-20.9 km·h-1, 21-23.9 km·h-1,>24 km·h-1, peak speed, accelerations (Acc) and decelerations (Dec)>1.0 m·s-2 and>2.5 m·s-2 and rate of perceived exertion (RPE) were recorded. Before and after each TG, countermovement-jump (CMJ), 15- (S15) and 30 m (S30) speed tests were assessed. TG3vs2 showed greater DC and Dec>1.0 m·s-2 than TG2vs1, and DC, DC 18.0-23.9 km·h-1, Acc>1.0 m·s-2 and Dec>2.5 m·s-2 than TG1vs1 (p<0.01). TG2vs1 achieved higher DC, DC 18.0-23.9 km·h-1, and Acc>2.5 m·s-2 (p<0.01) but lower peak speed (p=0.02) and RPE (p=0.02) than TG1vs1. Post-intervention, TG1vs1 showed lower CMJ and higher S15 (p=0.02), while TG3vs2, showed improvements in CMJ (p<0.01). The three tasks showed large variations for DC>24 km·h-1, Acc>1.0 m·s-2, Dec>1.0 m·s-2 and Dec>2.5 m·s-2. The load of TGs is sensitive to their player composition.

The aim of this study was to analyze the differences in internal and external load during Soccer-7 and Soccer-11, comparing positional requirements and neuromuscular fatigue in both modalities. Twenty-four young soccer players were monitored in Soccer-7 and Soccer-11 matches using global positioning systems. Total distance covered (TD), distance covered at high speed (HSR), distance covered at very high speed (VHSR), peak speed, accelerations (Acc) and decelerations (Dec) and rate of perceived exertion (RPE) were recorded differentiating between central backs (CB), midfielders (MF), external players (EX) and forwards (FW). Neuromuscular fatigue were assessed using a jump test. During Soccer-11, players showed significantly higher TD, HSR and VHSR, with low Acc and greater RPE compared with Soccer-7. During Soccer-11, all positions recorded significantly greater TD, distance at HSR and at VHSR than Soccer-7. In terms of playing position, CB, MF and FW achieved significantly higher Peak Speed during Soccer-1, but there was no difference for EX. During Soccer-7 all positions performed significantly higher numbers of Acc. Although the Soccer-7 modality is considered an optimal format for the development of young soccer players, there is a significant difference in match running activity for all playing positions with respect to the Soccer-11 format.

Studies have reported increased intestinal permeability in multiple sclerosis (MS) patients and its mouse model experimental autoimmune encephalomyelitis (EAE). However, the mechanisms driving increased intestinal permeability that in turn exacerbate neuroinflammation during EAE remain unclear. Here we showed that vancomycin preserved the integrity of the intestinal barrier, while also suppressing gut trypsin activity, enhancing the relative abundance of specific Lactobacilli and ameliorating disease during EAE. Furthermore, Lactobacilli enriched in the gut of vancomycin-treated EAE mice at day 3 post immunization negatively correlated with gut trypsin activity and EAE severity. In untreated EAE mice, we observed increased intestinal permeability and increased intestinal protease activated receptor 2 (PAR2) expression at day 3 post immunization. Prior studies have shown that trypsin increases intestinal permeability by activating PAR2. Our results suggest that the interaction between intestinal PAR2 and trypsin may be a key modulator of intestinal permeability and disease severity during EAE.

The aims of this study were to compare the influence of transition game (TG) size on the external and internal loads of young professional soccer players and to describe the high-speed profile of these drills in response to pitch dimensions. Eighteen young professional soccer players (age: 16.1 ± 0.3 years; height: 178.3 ± 5.4 cm; weight: 70.1 ± 6.2 kg) performed a 3vs2 TG on pitches measuring 40 × 30 m (TG30), 40 × 50 m (TG50) and 40 × 70 m (TG70) m. Distance covered (DC); accelerations-decelerations above 1.0 m · s-2 and 2.5 m · s-2; rate of perceived exertion (RPE); maximal heart rate and time above 90%; DC at 18.0 to 21.0 km · h-1 (DC 18-20.9 km · h-1); DC at 21.0 to 23.9 km · h-1 (DC 21-23.9 km · h-1); DC above 24.0 km · h-1 (DC > 24 km · h-1); and peak speed and sprint profile (duration, distance and maximal speed) were measured. TG30 achieved lower DC, DC above 18 km · h-1, peak speed, sprint distance and RPE than TG50 and TG70 (p < 0.01 and p < 0.05) and lower sprint duration and maximal speed sprint than TG70 (p < 0.01). TG30 and TG50 achieved higher Acc > 1.0 and > 2.5 m · s-2 respectively than TG70 (p < 0.05). TG70 showed greater DC above 21 km · h-1, peak speed, sprint distance and maximal speed sprint than TG50 (p < 0.01). Soccer coaches should use larger TGs to overload variables related to high speed and sprint demands during training and smaller TG formats to stimulate the accelerations of the soccer players.

Rationale: Chronic obstructive pulmonary disease (COPD) is associated with high morbidity, mortality, and healthcare costs. Cigarette smoke is a causative factor; however, not all heavy smokers develop COPD. Microbial colonization and infections are contributing factors to disease progression in advanced stages. Objectives: We investigated whether lower airway dysbiosis occurs in mild-to-moderate COPD and analyzed possible mechanistic contributions to COPD pathogenesis. Methods: We recruited 57 patients with a >10 pack-year smoking history: 26 had physiological evidence of COPD, and 31 had normal lung function (smoker control subjects). Bronchoscopy sampled the upper airways, lower airways, and environmental background. Samples were analyzed by 16S rRNA gene sequencing, whole genome, RNA metatranscriptome, and host RNA transcriptome. A preclinical mouse model was used to evaluate the contributions of cigarette smoke and dysbiosis on lower airway inflammatory injury. Measurements and Main Results: Compared with smoker control subjects, microbiome analyses showed that the lower airways of subjects with COPD were enriched with common oral commensals. The lower airway host transcriptomics demonstrated differences in markers of inflammation and tumorigenesis, such as upregulation of IL-17, IL-6, ERK/MAPK, PI3K, MUC1, and MUC4 in mild-to-moderate COPD. Finally, in a preclinical murine model exposed to cigarette smoke, lower airway dysbiosis with common oral commensals augments the inflammatory injury, revealing transcriptomic signatures similar to those observed in human subjects with COPD. Conclusions: Lower airway dysbiosis in the setting of smoke exposure contributes to inflammatory injury early in COPD. Targeting the lower airway microbiome in combination with smoking cessation may be of potential therapeutic relevance.

BACKGROUND: There is an urgent need to identify biomarkers for advanced adenoma, an important precursor of colorectal cancer (CRC). We aimed to determine alterations in ileal juice bile acids associated with colorectal advanced adenoma. METHODS: We quantified a comprehensive panel of primary and secondary bile acids and their conjugates using an ultraperformance liquid chromatography triple-quadrupole mass spectrometric assay in ileal juice collected at colonoscopy from 46 study subjects (i.e., 14 biopsy-confirmed advanced adenomas and 32 controls free of adenoma or cancer). Using analysis of covariance (ANCOVA), we examined the differences in bile acid concentrations by disease status, adjusting for age, sex, body mass index, smoking status and type 2 diabetes. RESULTS: The concentrations of hyodeoxycholic acid (HCA) species in ileal juice of the advanced adenoma patients (geometric mean = 4501.9 nM) were significantly higher than those of controls (geometric mean = 1292.3 nM, p = 0.001). The relative abundance of ursodeoxycholic acid (UDCA) in total bile acids was significantly reduced in cases than controls (0.73% in cases vs. 1.33% in controls; p = 0.046). No significant difference between cases and controls was observed for concentrations of total or specific primary bile acids (i.e., cholic acid (CA), chenodeoxycholic acid (CDCA) and their glycine- and taurine-conjugates) and total and specific major secondary bile acids (i.e., deoxycholic acid and lithocholic acid). CONCLUSIONS: Colorectal advanced adenoma was associated with altered bile acids in ileal juice. The HCA species may promote the development of colorectal advanced adenoma, whereas gut microbiota responsible for the conversion of CDCA to UDCA may protect against it. Our findings have important implications for the use of bile acids as biomarkers in early detection of colorectal cancer.

Although increasing evidence links microbial dysbiosis with the risk for psychiatric symptoms through the microbiome-gut-brain axis (MGBA), the specific mechanisms remain poorly characterized. In a diagnostically heterogeneous group of treated psychiatric cases and nonpsychiatric controls, we characterized the gut and oral microbiome, plasma cytokines, and hippocampal inflammatory processes via proton magnetic resonance spectroscopic imaging (1H-MRSI). Using a transdiagnostic approach, these data were examined in association with schizophrenia-related symptoms measured by the Positive and Negative Syndrome Scale (PANSS). Psychiatric cases had significantly greater heterogeneity of gut alpha diversity and an enrichment of pathogenic taxa, like Veillonella and Prevotella, in the oral microbiome, which was an accurate classifier of phenotype. Cases exhibited significantly greater positive, negative, and general PANSS scores that uniquely correlated with bacterial taxa. Strong, positive correlations of bacterial taxa were also found with cytokines and hippocampal gliosis, dysmyelination, and excitatory neurotransmission. This pilot study supports the hypothesis that the MGBA influences psychiatric symptomatology in a transdiagnostic manner. The relative importance of the oral microbiome in peripheral and hippocampal inflammatory pathways was highlighted, suggesting opportunities for probiotics and oral health to diagnose and treat psychiatric conditions.

The microbiomes of cesarean-born infants differ from vaginally delivered infants and are associated with increased disease risks. Vaginal microbiota transfer (VMT) to newborns may reverse C-section-related microbiome disturbances. Here, we evaluated the effect of VMT by exposing newborns to maternal vaginal fluids and assessing neurodevelopment, as well as the fecal microbiota and metabolome. Sixty-eight cesarean-delivered infants were randomly assigned a VMT or saline gauze intervention immediately after delivery in a triple-blind manner (ChiCTR2000031326). Adverse events were not significantly different between the two groups. Infant neurodevelopment, as measured by the Ages and Stages Questionnaire (ASQ-3) score at 6 months, was significantly higher with VMT than saline. VMT significantly accelerated gut microbiota maturation and regulated levels of certain fecal metabolites and metabolic functions, including carbohydrate, energy, and amino acid metabolisms, within 42 days after birth. Overall, VMT is likely safe and may partially normalize neurodevelopment and the fecal microbiome in cesarean-delivered infants.

The aim of this study was to evaluate the effects of different time periods on the internal and external loads of soccer players during small-sided games (SSGs). Seventeen young soccer players performed a SSG of 5 vs. 5 + 5 with 2 floaters, where two teams had possession of the ball, and one had to recover it. With established periods of 30 s (SSG30), 1 min (SSG1) or 2 min (SSG2), teams participated in a defensive role for these periods of time. Total distance covered, moderate speed running distance, high speed running distance, sprint running distance, accelerations, decelerations and Player Load were monitored using global positioning systems devices. The maximal heart rate and modified training impulse were monitored using heart rate monitors. The rating of perceived exertion (RPE) was also measured. Data showed a small increase between SSG30 and SSG1 in Player Load (ES = -0.35; p < 0.01), and a small increase in high speed running (ES = -0.41; p < 0.05) and sprinting (ES = -0.47; p < 0.01) between SSG30 and SSG2. Also, SSG1 showed a small increase in sprinting (ES = -0.57; p < 0.01) and accelerations (ES = -0.37; p < 0.05) with respect to SSG2. In addition, SSG2 showed a small increase in the RPE with respect to SSG30 (ES = 0.46; p < 0.05). The results indicate that shorter defensive periods in SSGs increased high speed running, while longer defensive periods promoted greater perceived exertion. Manipulation of the duration of defensive periods in SSGs is a variable that should be considered in soccer training.

The increasing demand for electrical energy storage makes it essential to explore alternative battery chemistries that overcome the energy-density limitations of the current state-of-the-art lithium-ion batteries. In this scenario, lithium-sulfur batteries (LSBs) stand out due to the low cost, high theoretical capacity, and sustainability of sulfur. However, this battery technology presents several intrinsic limitations that need to be addressed in order to definitively achieve its commercialization. Herein, we report the fruitfulness of three different formulations using well-selected functional carbonaceous additives for sulfur cathode development, an in-house synthesized graphene-based porous carbon (ResFArGO), and a mixture of commercially available conductive carbons (CAs), as a facile and scalable strategy for the development of high-performing LSBs. The additives clearly improve the electrochemical properties of the sulfur electrodes due to an electronic conductivity enhancement, leading to an outstanding C-rate response with a remarkable capacity of 2 mA h cm-2 at 1C and superb capacities of 4.3, 4.0, and 3.6 mA h cm-2 at C/10 for ResFArGO10, ResFArGO5, and CAs, respectively. Moreover, in the case of ResFArGO, the presence of oxygen functional groups enables the development of compact high sulfur loading cathodes (>4 mgS cm-2) with a great ability to trap the soluble lithium polysulfides. Notably, the scalability of our system was further demonstrated by the assembly of prototype pouch cells delivering excellent capacities of 90 mA h (ResFArGO10 cell) and 70 mA h (ResFArGO5 and CAs cell) at C/10.

Adolescents around the world do not engage in sufficient physical activity and the Spanish context is no exception. Understanding the educational context as a complex system, school-based multi-level and multi-component interventions seem to be an effective strategy to reverse this trend. Moreover, a co-creational approach seems to facilitate the mobilization of community partnerships and the engagement of stakeholders in the intervention process. This study aims to describe the dissemination, implementation, and evaluation process of an effective school-based intervention program in another setting using the replicating effective programs framework and a co-participatory approach. This study will be conducted in two Spanish secondary schools located in the region of Aragon (experimental vs. control school) in a sample of adolescents in the second grade (13-14 years old). To evaluate the effectiveness, different health behaviors such as physical activity, sleep, sedentary time with screens, nutrition, and psychosocial variables will be quantitatively measured at baseline and after the implementation of the intervention. Qualitative methods will also be used to better understand the implementation process and the co-creation approach, as well as to provide insights into the sustainability of the intervention program. The current study has the potential to provide strong information about the dissemination, implementation, and evaluation process of school-based programs to promote healthy behaviors among adolescents.

RESUMO

OBJECTIVE: To provide practical recommendations for the comprehensive approach of people with type 2 diabetes according to evidence-based medicine. PARTICIPANTS: Members of the Diabetes Knowledge Area of the Spanish Society of Endocrinology and Nutrition. METHODS: The recommendations were formulated according to the degrees of evidence of the Standards of Medical Care in Diabetes-2022. After reviewing the available evidence and formulating recommendations by the authors of each section, several rounds of comments were developed incorporating the contributions and voting on controversial points. Finally, the final document was sent to the rest of the members of the area for review and incorporation of contributions, to finally carry out the same process with the members of the Spanish Society of Endocrinology and Nutrition Board of Directors. CONCLUSIONS: The document establishes practical recommendations based on the latest available evidence for the management of people with type 2 diabetes.

A central tenet of many theories of emotion is that emotional states are accompanied by distinct patterns of autonomic activity. However, experimental studies of coherence between subjective and autonomic responses during emotional states provide little evidence of coherence. Crucially, previous studies investigating coherence have either adopted univariate approaches or made limited use of multivariate analytic approaches by investigating subjective and autonomic responses separately. The current study addressed this question using a multivariate dimensional approach to build a common autonomic-subjective affective space incorporating subjective responses and three different autonomic signals (heart rate, skin conductance response, and pupil diameter), measured during an emotion-inducing task, in 51 participants. Results showed that autonomic and subjective responses could be adequately described in a two-dimensional affective space. The first dimension included contributions from subjective and autonomic responses, indicating coherence, while contributions to the second dimension were almost exclusively of autonomic covariance. Thus, while there was a degree of coherence between autonomic and subjective emotional responses, there was substantial structure in autonomic responses that did not covary with subjective emotional experience. This study, therefore, contributes new insights into the relationship between subjective and autonomic emotional responses, and provides a framework for future multimodal emotion research, enabling both hypothesis- and data-driven testing.

This study examined the training load on professional soccer players during transition games performed with different bout durations and their effects on speed and jump tests. Fourteen young soccer players played a transition game of different bout durations: 15 (TG15), 30 (TG30), and 60 (TG60) seconds. Total distance covered (DC), accelerations and decelerations above 1.0 and 2.5 m·s-2, rate of perceived exertion (RPE), maximal heart rate (HRmax) and above 90% (HR>90), distance covered between 18.0-20.9 km·h-1 (DC 18.0-20.9 km·h-1), 21.0-23.9 km·h-1 (DC 21.0-23.9 km·h-1), above 24.0 km·h-1 (DC>24.0 km·h-1), peak speed, sprint profile, sprint, and countermovement jump tests were recorded. TG15 obtained greater DC, DC>21.0 km·h-1, Player load, Acc>2.5 m·s-2 than TG30 and TG60 (p<0.01) and Acc<2.5 m·s-2, Dec>2.5 m·s-2 than TG60 (p<0.01). TG30 showed more HR>90 and RPE (p<0.01) than TG15, and DC, DC>18.0 km·h-1, Player load, Acc>2.5 m·s-2, Dec>2.5 m·s-2, HR>90 and RPE than TG60 (p<0.01 and<0.05). Transition games showed lower sprint and jump results after the intervention (p<0.01). Bout duration has been configured as an important constraint that influences the transition games and the performance of soccer players.

OBJECTIVES: To investigate the cutaneous microbiome spanning the entire psoriatic disease spectrum, and to evaluate distinguishing features of psoriasis (PsO) and psoriatic arthritis (PsA). METHODS: Skin swabs were collected from upper and lower extremities of healthy individuals and patients with PsO and PsA. Psoriatic patients contributed both lesional (L) and contralateral non-lesional (NL) samples. Microbiota were analysed using 16S rRNA sequencing. RESULTS: Compared with healthy skin, alpha diversity in psoriatic NL and L skin was significantly reduced (p<0.05) and samples clustered separately in plots of beta diversity (p<0.05). Kocuria and Cutibacterium were enriched in healthy subjects, while Staphylococcus was enriched in psoriatic disease. Microbe-microbe association networks revealed a higher degree of similarity between psoriatic NL and L skin compared with healthy skin despite the absence of clinically evident inflammation. Moreover, the relative abundance of Corynebacterium was higher in NL PsA samples compared with NL PsO samples (p<0.05), potentially serving as a biomarker for disease progression. CONCLUSIONS: These findings show differences in diversity, bacterial composition and microbe-microbe interactions between healthy and psoriatic skin, both L and NL. We further identified bacterial biomarkers that differentiate disease phenotypes, which could potentially aid in predicting the transition from PsO to PsA.
